High Throughput Screening Services

  • Vector cloning: we subclone your gene of interest into our portfolio of expression vectors, covering a representative set of fusion constructs.
    • Subsequent expression optimization of the resulting plasmids in a set of E. coli strains is then performed to maximize the amount of soluble, fully-folded, active protein.
    • Established cell lines are generated with your wild-type or mutated gene of interest.
  • Protein purification: scale-up of the optimal expression condition is followed by scouting of different lysis methods, chromatographic steps and a buffer formulation screen, to push the pure protein yield to the milligram level, so that it is ready to go for your biophysical experiments and your target-based screens.

Quality Controls: We perform QC of each screening plate automatically on signal/background ratio and Z´ score calculations, considering the best plate layout for your assay; a statistically optimal number of repeats are performed for the internal positive and negative controls, and/or a dose-response curve of relevant validated inhibitors when available. Genedata Screener® software and in-house applications help us perform the analysis, visualizations, and management of screening data.
